DETECTION OF THE IS6110 GENE OF MYCOBACTERIUM TUBERCULOSIS IN THE SERUM OF HOUSEHOLD CONTACTS OF TUBERCULOSIS PATIENTS USING OF POLYMERASE CHAIN REACTION (PCR) METHODE

Abstrak
Tuberculosis is one of the diseases with the highest mortality rates in the world. Tuberculosis is an infectious disease that remains a global health problem, especially in developing countries like Indonesia. Tuberculosis transmission is closely related to close contact in the household environment. Early detection of tuberculosis infection, especially in high-risk individuals, is crucial. One currently developing approach is molecular diagnostics, which is an examination that detects the presence of genetic material from Mycobacterium tuberculosis. The use of the Polymerase Chain Reaction method allows for rapid, sensitive, and specific amplification of specific DNA fragments from tuberculosis bacteria. The IS6110 gene is a specific DNA sequence from Mycobacterium tuberculosis that can be used as a molecular marker in the early detection of latent Tuberculosis infection. This study aims to detect the presence of the IS6110 gene in the serum of individuals who have a history of household contact with active tuberculosis patients, using the Polymerase Chain Reaction method. This study used a descriptive experimental design with a molecular laboratory approach. A total of 17 serum samples from household contacts were collected and analyzed using PCR to amplify the IS6110 gene. The results showed that 1 of 17 samples (5.88%) was detected positive for the IS6110 gene, while the other 16 samples (94.12%) showed negative results.
